Alzheimer’s disease (AD) accounts for about 70% of all dementias, and approximately 10 million new cases are diagnosed each year worldwide, with the number of new cases expected to reach 76 million by 2030 and 8 million in Japan, where the population is aging. There is no effective treatment for AD, and it is still a major social problem that requires supportive care from family members and caregivers.
Although the causes of AD are largely unknown, the pathological features of the disease are the accumulation of amyloid-β protein in the brain, synaptic dysfunction, and a decrease in the number of nerve cells, which eventually leads to brain atrophy. Brain atrophy begins near the hippocampus and eventually spreads to the entire brain. Symptoms begin with short-term memory impairment, such as inability to remember phone numbers, and eventually the entire cognitive function disappears.
The leading hypotheses for the etiology of the disease are a decrease in acetylcholine and an excess of glutamate. Cholinesterase inhibitors and glutamate antagonists have been developed and used as approved drugs to alleviate symptoms, but their efficacy is unknown.
SAISEIKEN Co. Ltd has developed an ultrasound culture method (patent number:7125818) and succeeded in producing a highly functional conditioned medium culture ( CM ) derived from deciduous teeth pulp stem cells. This new CM contains many bioactive substances that are effective for nerve regeneration. In fact, when the highly functional CM was used in 20 AD patients, clear therapeutic effects were shown after 8 weeks of intranasal administration. The paper describes these details.
“ Ultrasound-Mediated Augmented Conditioned Medium Derived From Dental Pulp Stem Cells Ameliorates Alzheimer’s Disease ”
Mesenchymal stem cells (MSCs) are a popular platform for regenerative medicine due to their ability to home to damaged organs and secrete molecules that spur cell growth and suppress inflammation. However, there remains a need to optimize their therapeutic effect for clinical translation.
One such strategy is the use of ultrasound. Ultrasound can be applied to MSCs to enhance their ability to secrete regenerative molecules and homing ability.
On the other hand, there have been significant developments in stem-cell-free therapy, which does not use stem cells. We have found that conditioned medium (CM) obtained by culturing stem cells from human exfoliated deciduous teeth (SHED) had effects of restoring memory in AD mice, inducing neurite outgrowth of nervous system cells, and inhibiting apoptosis.
In CM therapy, the creation of highly functional CM is the key to successful treatment. Ultrasound culture is the most appropriate method for creating highly functional CM because it can easily control the differentiation of stem cells.
In this study, I report the results of treatment of Alzheimer’s disease using ultrasound-mediated augmented CM derived from SHED and its remarkable improvement for the symptoms in Alzheimer’s disease. ( submitted to “ Cytotherapy ” in 2022.11.23)
